How to Document an Experiment for Reliable Replication

Clear, structured records let any competent researcher repeat your work with confidence.

Start every record with a concise title and a brief objective that states the hypothesis or question being tested. Include the date, the name of the person performing the work, and a unique experiment identifier so the file can be located later.

List all reagents, consumables, and equipment with catalog numbers, lot numbers, and any special preparation steps. Record concentrations, volumes, and the order of addition in a step-by-step format. For instruments, note model, firmware version, and all relevant settings such as temperature, speed, and detection parameters.

Capture raw data in its original form and annotate any observations made during the run. When processing data, describe the software, version, and analysis scripts used, and attach the exact code or calculation steps. Store files in a logical folder hierarchy and back them up on a secure server.

Use a lab notebook-paper or electronic-that timestamps each entry and allows for version control. Before finalizing the protocol, have a colleague review the documentation for completeness and clarity. A short checklist at the end of the record can confirm that all critical details have been captured.

When the experiment is complete, archive the protocol together with the data set and any supplementary files. This practice not only facilitates replication by others but also helps you troubleshoot future variations of the same assay.
