Start each record with a clear objective and hypothesis. State the scientific question, the expected outcome, and any constraints such as time or equipment limits. This framing helps anyone reading the protocol understand why each step was chosen.
List all reagents, consumables, and equipment with lot numbers, catalog identifiers, and calibration dates. Include concentrations, volumes, and preparation methods for solutions. When using instruments, note model numbers, firmware versions, and specific settings like temperature, speed, or voltage.
Describe the procedure step by step in the order performed, using precise verbs and measurable actions. Record timing, order of addition, mixing speeds, and any pauses. If a step required visual confirmation (e.g., color change, precipitate formation), note the criteria used to judge completion.
Document any deviations from the planned protocol, unexpected observations, and troubleshooting actions taken. Include raw data files, screenshots, or instrument logs as attachments, and reference them in the text. This creates a complete audit trail for later analysis.
Finally, store the record in an electronic lab notebook or a version-controlled repository with a descriptive file name and metadata tags for project, date, and researcher. Share the file with collaborators and, when appropriate, deposit it in a public data repository to enable full reproducibility.
