How to Document an Experiment for Reproducibility

Clear, concise record-keeping ensures others can repeat your work.

Start every entry with the date, project identifier, and a brief objective. State the hypothesis or question you are testing in one sentence, then list all reagents, cell lines, and equipment with catalogue numbers or lot identifiers. This creates a traceable chain from concept to material.

Describe the protocol step-by-step in the order you performed it. Include volumes, concentrations, incubation times, temperatures, and any deviations from standard methods. Record observations in real time - color changes, precipitates, instrument readouts - and note any unexpected events. Attach a copy of any instrument method files or software settings that influence the outcome.

When you generate data, save raw files in a dedicated folder with a consistent naming scheme that reflects the experiment date and sample ID. Use a spreadsheet or lab-information system to log processed results, calculations, and statistical analyses. Include the version of any analysis script and a brief comment on the parameters used.

Before you close the notebook, add a concise summary that highlights what worked, what didn’t, and any follow-up steps. Attach scanned copies of critical documents such as certificates of analysis or calibration logs. Finally, review the entry against a reproducibility checklist - complete material list, full protocol, raw data availability, and clear conclusions - to ensure another researcher can repeat the experiment without ambiguity.
