How to Document Experiments for Reliable Reproducibility

A concise guide to recording methods so others can repeat your work

Begin by outlining the experimental goal and the exact steps you plan to follow before you start any bench work. A brief protocol written in plain language helps you stay focused and provides a clear template for later documentation. Include the purpose, hypothesis, and any critical assumptions at the top of the page.

Use a bound lab notebook or an approved electronic notebook to capture every detail as the experiment progresses. Record the date, sample identifiers, reagent lot numbers, concentrations, volumes, and equipment settings. Note any deviations from the original plan, unexpected observations, and the time each step was performed. Consistent handwriting or a standardized digital entry format makes the record easy to read.

Save raw data files with descriptive filenames that include the experiment date, sample ID, and instrument used. Attach a short read-me file that lists file formats, software versions, and any processing steps applied. Store the files in a backed-up folder that mirrors the notebook entry, and consider uploading the complete set to a shared repository for peer access.

Before closing the notebook entry, review the entire record for completeness and clarity. Ask a colleague to read the documentation and confirm that they could repeat the experiment based solely on your notes. This final check catches missing information and reinforces good documentation habits.
