How to Reconstitute a Lyophilized Peptide Safely

Step-by-step guidance for restoring lyophilized peptides while preserving activity and purity.

Begin by reviewing the peptide’s datasheet for recommended solvent, pH range, and concentration limits. Use high-purity water or the specified buffer, and pre-warm it to room temperature unless the peptide is known to be heat-sensitive. Add the solvent slowly to the vial, aiming for a final volume that yields the desired stock concentration while staying within the peptide’s solubility limits.

Mix gently by swirling the vial or using a low-speed vortex for a few seconds. Avoid vigorous shaking, pipetting up and down, or sonication, as these actions can introduce shear stress or promote aggregation. If the peptide does not dissolve completely, allow the solution to sit at 4 °C for a short period, then repeat gentle mixing.

Once fully dissolved, aliquot the solution into low-binding microcentrifuge tubes to minimize adsorption to plastic surfaces. Store aliquots at the temperature recommended by the supplier, typically -20 °C or -80 °C, and protect them from repeated freeze-thaw cycles. If long-term storage is needed, consider adding a small amount of stabilizing agent such as glycerol, but only if compatible with downstream assays.

Before using the reconstituted peptide, verify its concentration by measuring absorbance at 280 nm or using a suitable assay. Check for visible precipitation or cloudiness; if present, filter the solution through a low-protein-binding syringe filter (0.22 µm) to remove particulates without exposing the peptide to harsh conditions. Record all reconstitution details in your lab notebook for reproducibility.

Finally, label each aliquot with the peptide name, concentration, reconstitution date, and storage conditions. Dispose of any unused solvent according to your institution’s safety protocols, and clean the work area to prevent cross-contamination with other peptides or reagents.
