Selecting the Right Syringe for Precise Microliter Volumes

Practical guidance for choosing a syringe that delivers reliable measurements in the 1-10 µL range.

When measuring volumes below 10 µL, the syringe’s tolerance becomes the dominant source of error. Look for syringes that specify a tolerance of ±1 % or better for the intended volume range, and verify that the manufacturer provides a calibration certificate.

Barrel size and tip type are closely linked to accuracy. For volumes under 5 µL, a 10 µL Hamilton or equivalent glass syringe with a Luer slip tip offers the most consistent dead-space. Plastic syringes can be used for larger ranges (5-20 µL) but be aware that they may expand slightly with temperature changes.

To minimize systematic error, pre-wet the syringe by aspirating and dispensing the solvent several times before taking a measurement. This reduces surface tension effects and eliminates air bubbles that can skew the readout. Perform all measurements at a stable room temperature, as temperature shifts can alter both liquid density and syringe dimensions.

Regular maintenance is essential for continued precision. Inspect the plunger and barrel for scratches or wear, clean the tip with appropriate solvent, and store the syringe vertically in a dust-free container. Periodically re-calibrate against a gravimetric standard to confirm that the syringe remains within specification.
