Endotoxin and bioburden are two distinct contaminants that can affect in-vitro experiments. Endotoxin is a lipopolysaccharide component of the outer membrane of Gram-negative bacteria, while bioburden refers to the total viable microbial load present in a sample.
The most common endotoxin test is the Limulus Amebocyte Lysate (LAL) assay; recombinant factor C kits provide a non-animal alternative. Typical workflow includes reconstituting the kit reagents, preparing the sample in endotoxin-free water, adding spiked controls, measuring optical density or kinetic absorbance, and calculating endotoxin units per milliliter using the standard curve.
Bioburden screening usually follows a microbial limit test. Samples are inoculated into a suitable growth medium and incubated under defined conditions; colony forming units are then counted. For low-level bioburden, membrane filtration followed by incubation on agar plates can improve detection sensitivity.
Good laboratory practice includes using endotoxin-free consumables, avoiding glassware that can adsorb endotoxin, and performing a pre-filter step when high particulate loads are expected. Always include positive and negative controls, record raw data, and retain documentation for audit purposes.
Interpretation of results should be based on pre-defined acceptance criteria. Results that exceed the limit require additional purification steps or disposal of the batch.
