How to accurately determine the usable peptide amount in a vial by accounting for counterions, water and other non-peptide mass.
Gross vial mass includes everything inside the container - the peptide, any counter-ions, residual solvents, and water that may be bound to the lyophilized cake. Net peptide content is the mass of the peptide itself and is the figure you need for stoichiometric calculations and stock solution preparation.
To calculate net peptide mass, start with the gross mass measured on an analytical balance after subtracting the empty vial weight. Then subtract the mass contributed by counter-ions (e.g., TFA, acetate) using the known stoichiometry and molecular weight of each ion. If the certificate of analysis lists a moisture content, subtract that percentage of the gross mass as water.
A practical workflow is: (1) tare the balance with an empty vial; (2) weigh the sealed vial and record the gross mass; (3) obtain the vial’s empty weight from the supplier or measure an identical empty vial; (4) apply the counter-ion and moisture corrections. The resulting net mass can be rounded to the nearest microgram for precise pipetting.
Using the net peptide mass ensures that your reconstitution calculations are accurate. For example, if you need a 1 mg mL stock, dissolve the net peptide mass (not the gross mass) in the appropriate volume of solvent. Overlooking counter-ion mass can lead to lower than intended concentrations, affecting downstream assays.
Tips for reliable results: store vials at the recommended temperature, avoid condensation when opening, and re-weigh the vial after any moisture-sensitive handling. Document all measured and calculated values in your lab notebook for traceability.
