Sequence and non-standard residues
The sequence is H-Aib-His-D-2Nal-D-Phe-Lys-NH2. Three features separate it from a peptide built only from proteinogenic L-amino acids. The N-terminal residue is Aib (α-aminoisobutyric acid, or 2-methylalanine), which carries two methyl groups on the α-carbon. The third residue is D-2-naphthylalanine, a D-configured amino acid with a naphthyl side chain. The fourth is D-phenylalanine. The C-terminus is a primary amide rather than a free carboxylic acid.
These features matter for analysis. The amide and the D-residues change retention behaviour and fragmentation compared with a free-acid, all-L analogue. A reference standard and a method must therefore be specific to this exact structure.
Formula and expected masses
The molecular formula is C38H49N9O5. The average molecular weight is about 711.9 g/mol and the monoisotopic mass is 711.3857 Da. In positive electrospray ionisation the singly protonated ion [M+H]+ is expected near m/z 712.393 and the doubly protonated ion [M+2H]2+ near m/z 356.700. The lysine side chain and the imidazole of histidine make multiply charged ions easy to form, so both species commonly appear in a spectrum.
The peptide was first described in the primary literature as a synthetic pentapeptide agonist of the growth hormone secretagogue receptor (PMID: 9849822). That paper is the usual starting point for the structural background.
Identity confirmation on a certificate
A certificate of analysis normally combines two orthogonal measurements. Reversed-phase HPLC with UV detection gives a chromatogram. Identity is supported when the main peak's retention time matches that of a reference standard run under the same conditions. Mass spectrometry gives the measured m/z, which should agree with the calculated values above within the tolerance of the instrument.
Each measurement has limits. A retention-time match shows only that the sample behaves like the standard under one set of conditions. A mass match confirms composition, not stereochemistry, because a diastereomer with the same formula has the same mass. Confirming the D-configuration at positions 3 and 4 needs a chiral method or a comparison of fragment intensities against a standard. A routine certificate does not necessarily include this, so check which tests were actually run.
High-resolution tandem MS can go further. Nano-liquid chromatography coupled to an orbitrap analyser has been applied to small synthetic peptides in complex matrices, with accurate-mass precursor and product ions used for assignment (PMID: 22901302). The same approach applies to a neat research sample, where fragment ions can confirm the order of residues.
Reading the report
A certificate reports what one laboratory measured on one sample of one batch. Testing is performed by a third-party laboratory, which is named on each report. Vials carry no lot number, so the document is matched to the material through the Report ID or Laboratory ID printed on the certificate. Some batches may still be awaiting results.
When reading a report, check these items:
First, the method description: column, gradient, detection wavelength, and whether a reference standard was used. Second, the observed m/z values, compared against 712.39 and 356.70. Third, the date and the laboratory's name. Fourth, whether any peaks other than the main one are integrated, and how they are described. An area percentage from a UV trace is a ratio of detector responses under one method, not an absolute statement about composition.
Receptor pharmacology as bench measurement
Receptor pharmacology for this compound belongs in vitro: radioligand competition binding at the GHS-R1a receptor, and cell-based functional readouts such as calcium mobilisation or second-messenger assays in transfected cell lines, reported as Ki or EC50 values. These numbers depend on cell line, receptor expression level, assay buffer and incubation time, so values from different laboratories are not interchangeable. They are also not part of a standard identity certificate. Anyone running such assays should qualify the material in their own system against a suitable control ligand.
Stability and bench handling
Lyophilised peptide is generally kept sealed, cold, dry and protected from light. Vials should reach room temperature before opening so that condensation does not wet the powder. Weigh quickly, because lyophilised peptides take up moisture.
For assay work, a stock solution is prepared at the bench in a solvent compatible with the downstream system, then divided into small aliquots to avoid repeated freeze-thaw cycles. Solutions are usually less stable than the dry powder, so how long a stock remains usable should be established by the laboratory, for example by re-running HPLC on an aged aliquot against a freshly prepared one.
Degradation can be followed by LC-MS. Published work on the metabolism of this peptide class has described how mass spectrometry is used to characterise truncated and modified forms (PMID: 23101768). The same logic applies to stored stocks: new peaks with masses lower than 711.39 Da may indicate cleavage products, while +16 Da shifts suggest oxidation.
