ARA-290, also called cibinetide, is an eleven-residue synthetic peptide built from the aqueous-facing residues of helix B of human erythropoietin. It is linear, and its first residue is pyroglutamate rather than a free amine.

The sequence is Pyr-Glu-Gln-Leu-Glu-Arg-Ala-Leu-Asn-Ser-Ser, carried as the free acid. One point about that sequence is worth stating plainly, because it is easy to get wrong: it is not a contiguous stretch of erythropoietin. Brines and colleagues, reporting the design in PNAS in 2008, assembled it from residues that sit together on the aqueous face of helix B in the folded protein but are spread across the linear chain, so searching the erythropoietin sequence in UniProt P01588 for these eleven letters returns nothing.

The N-terminal pyroglutamate is a lactam formed from glutamate, which is why the formula registered at PubChem CID 91810664 and FDA GSRS is the free-acid undecapeptide minus ammonia. There is no acetylation, no amidation, no non-standard residue other than that pyroglutamate, and no metal coordination reported.

The molecule contains no cysteine and no sulfur atom of any kind, so there is no disulfide to form, scramble or reduce. It also contains no tryptophan, tyrosine or phenylalanine, which means there is no aromatic chromophore and absorbance at 280 nm returns nothing useful; concentration has to come from peptide-bond absorbance in the low ultraviolet, amino-acid analysis or a mass-based method.

The reported target is a heteromeric receptor complex rather than a single protein: the erythropoietin receptor EPOR (UniProt P19235) together with the shared beta-common subunit CD131, the product of CSF2RB (UniProt P32927), which Brines and colleagues named the innate repair receptor. No experimental structure of this peptide bound to that complex exists, so the contact residues are not established.

Several things commonly printed beside this compound are not established in the primary literature and are omitted here rather than estimated: there is no published solution-conformation study by circular dichroism or NMR, no quantitative solubility figure, no forced-degradation study, and no registry value for the counter-ion of supplied material. A proposed interaction with TRPV1 rests on a single secondary report and is not treated here as characterised.
